Construction and Use
The kit comprises a sterilized plastic or glass bottle containing a dry powder medium that is fully soluble in water, forming a dark amber solution; this solution turns black in the presence of H2S-producing organisms at ambient temperatures between 25°C and 44°C (preferably 35-37°C), over an observation period of 24 to 48 hours, with no incubator needed within that temperature range. It can be used with water from any source, including chlorinated water.
Interpreting Results
As a microbiological water testing kit, it is not a replacement for the conventional Presumptive Coliform test and other laboratory-based tests, and results should be treated with caution since a false positive is more likely than a false negative; in groundwater, naturally occurring sulphides may also trigger a positive H2S result rather than true faecal contamination, an error that errs on the side of safety. The kit is well suited to educational and motivational field campaigns promoting water sanitation and hygiene. Where the ambient temperature is below 25°C, the kit should be used with an incubator at an optimal 35-37°C, and kits should always be stored, boxed, in a cool, dry place away from sunlight, at a room temperature of no more than 30°C.
